Skrócony opis:

Zarodek ptasi stanowi wygodny model do szeroko pojętych badań biologicznych. Wynika to głownie z faktu, że rozwija się on poza organizmem matki, przebywając jednocześnie w stabilnym pod względem chemicznym środowisku wnętrza jaja i przy ograniczonym, błonami płodowymi oraz skorupą wapienną, kontakcie ze środowiskiem zewnętrznym.

Badania z użyciem zarodków można ogólnie podzielić na: 1) bezinwazyjne, kiedy skorupa jaja i błony podskorupowe nie zostają naruszone oraz 2) inwazyjne, wiążące się z ingerencją w strukturę jaja. Do pierwszej grupy można zaliczyć np. badania nad wpływem czynników środowiskowych na embriogenezę takich jak pola elektromagnetyczne, stres cieplny czy niedotlenienie. Do drugiej grupy należą badania polegające na podaniu in ovo substancji chemicznych lub immunologicznych oraz eksperymenty mikrochirurgiczne i biotechnologiczne, związane z ingerencją w tkanki zarodka.

Avian embryos are easy to breed, inexpensive and relatively resistant to “in ovo” manipulation. In practical poultry production, the fact that embryos are tolerant to egg structure manipulation was used to develop a method for in ovo vaccination of embryos [. Today, this method is successfully used to immunize chickens and turkeys against diseases such as Gumboro disease (infectious bursal disease, IBD) and Marek’s disease (MD). For the reasons mentioned above, avian embryos are a recognized model (in ovo model) in embryological , genetic , endocrinological , microbiological , immunological, toxicological, pharmacological and medical research. However, some reservation is needed because, due to phylogenetic differences, the results obtained with the in ovo model should be treated with caution with reference to mammals and humans.

In terms of animal science, veterinary and toxicological studies, it is highly significant that in ovo development is free of the ontogenic biochemical effect of the mother, which is the case with in utero development in mammals. In the avian egg, due to the lack of placenta, all substances can only be deposited until shell formation . At the same time, the egg’s chemical composition is extremely conservative as regards the content of many components (e.g. amino acids) and can be modified through mother’s nutrition only for some of them (e.g. vitamins and minerals). For this reason, oral administration of the analysed substance to the layer does not guarantee that the assumed dose will reach the egg. This is made possible, however, through in ovo injection .

In ovo injection can generally be used not only in birds but in all oviparous animals such as reptiles, amphibians and fish . This procedure can be performed at different stages of embryo development, from diapause (prior to setting of eggs in an incubator) until the time preceding hatching This makes it possible to determine the action of a given substance depending on the developmental degree of organs, tissues and the endocrine and nervous systems. It should be pointed out, however, that embryo sensitivity to mechanical manipulation is inversely correlated with their stage of development. When studying the embryotoxic effects, in ovo injection should be performed as early during embryogenesis as possible. In turn, to investigate the effect of a given substance on the hatching process and/or postembryonic development, this procedure should be performed shortly before hatching (the period between embryonic days E17 and E18 is considered optimal in chickens). During this period, not only nutrients and their metabolites (carbohydrates, amino acids) but also vitamins, hormones, antioxidants, pro- and prebiotics can be administered in ovo for the purpose of stimulating the chick’s postembryonic development . Depending on the type of the experiment and the embryo’s developmental stage, a substance may be administered on the shell membrane, into the interior egg (albumen or yolk), into the yolk sac, amnion or albumen sac, or into the embryo body . During the period before hatching, substances can be dosed in the form of a liquid (solution, emulsion) and suspension, and also as a solid. In the latter case, the substance is injected into the air cell on the shell membrane. It is taken in by the chick during internal pipping. It should be noted that correct in ovo injection into the albumen sac (amniotic fluid) or air cell causes no damage to the embryo’s tissues and organs , but may have an effect on the rate of chick hatching. This is because making a hole in the shell depressurizes the air cell. As a result, external pipping is delayed with a concurrent increase in hatching synchronization. Injections performed during the first half of incubation were observed to have no effect on the hatching process
