Skrócony opis:

The aim of teaching: familiarization of master course students with the laboratory methods applied in animal science. During lectures basic concepts of concerning laboratory analytics and different laboratory methods, i.e. spectrophotometric, isotopic and immunologic applied in veterinary diagnostics and animal science will be presented. Following the course students should be familiar with principles of mono- and polyclonal antibody production and their application in different laboratory techniques such as radioimmunology, ELISA, Western blot, immunocytochemistry. During laboratories students will be carried out analyses with application of isotopes and antibodies for determination of hormones in plasma or serum of domestic animals. They will use immunocytochemistry for protein detection using specific mono- or polyclonal antibodies.

Lectures (15 h):

Instrumental methods, rules for the collection, preparation and storage of biological samples for laboratory tests. Methods of extraction of biological substances from animal tissues. The stages of the analytical process.

Spectrophotometric methods and their application to the determination of concentrations of organic and inorganic compounds in the blood plasma of animals.

Application of radioisotopes for veterinary diagnostic and in vivo and in vitro experiments in animal science (hormone kinetics, blood flow, uptake of a hormone, cell proliferation).

Mono- and polyclonal antibodies - their characteristics, and method of production.

Radioimmunoassay (RIA) - the principle of the method, cross-reactivity of antibodies, the test parallelism and recovery; Radioreceptorassay (RRA) – kinetics of radioligand assays, the Scatchard’s plot. Application of RIA and RRA methods in animal science.

Immunochemical methods (immunoassay techniques, the method ABC, fluorescent and chemiluminescent methods).

The use of antibodies in selected techniques, Part I: immunocytochemistry

Usage of antibodies in selected laboratory techniques, Part II: ELISA, Western blot, immunoprecipitation, immuno-PCR, EMSA.

Laboratories (15 h):

Collection of blood and tissue samples; radioimmunoassay method - determination of hormone levels in the blood of animals (5 h).

Immunocytochemistry (the location of apoptotic or proliferating cells on paraffin sections of tissue) (5 h).

Principles of different types of ELISA tests. Determination of hormones or proteins by ELISA (5 h).

The effect of education: understanding the concepts concerning application of modern analytical methods for determination of hormones and proteins in biological samples using isotopes and antibodies.

Kwnowledge:

The student defines instrumental methods; knows the rules for the collection, preparation and storage of biological samples for laboratory tests and the method of extraction of biological substances from animal tissues. explains the stages of the analytical process.

He/she specifies basic spectrophotometric methods and describes their application to determine concentrations of organic and inorganic compounds in blood plasma.

He/she describes and defines the basic concepts and issues of nuclear physics: atom, isotope, radioactive decay, types of radiation, the measurement of radiation, radioactive series, and has knowledge of the use of radioactive isotopes in animal science

The student explains the usage of isotopically labeled inorganic and organic compounds in the in vivo and in vitro studies.

The student explains ways and methods of mono-and polyclonal antibody production; describes and characterizes basic methods and laboratory techniques involving the use of antibodies and / or isotopes.

Skills:

He/she applies radioactive isotopes and antibodies for in in vitro and in vivo experiments; can perform some analysis using isotopically labeled substances and/or antibodies.

The student applies the method of immunocytochemistry in research and diagnostics of cells and tissues. He/she interprets the results of immunocytochemical analysis.

Student determines hormone concentration in blood plasma of domestic animals by radioimmunoassay (RIA).

She/he uses the ELISA method for protein and hormone determination in plasma samples of different animal species.

Social competences:

He/she can work in a team and manage a small team performing laboratory analyzes.

The student has a sense of responsibility, and the risks and consequences of genetic manipulation

She is aware of the importance of ethical principles in carrying out animal experiments, laboratory analyzes, and the correct interpretation of the test results.

Metody i kryteria oceniania:

The percentage scale for assessing educational results is defined as follows:

1. Fail (F): some more work required before the credit can be awarded; it is issued when during the course the student have obtained less than 55% of the required effects of at least from one of the three components of the educational results (K, S, SC)

2. Sufficient (E): performance meets the minimum criteria, it is issued when during the course the student have obtained at least 55% of the required effects of each components of the educational results (K, S, SC)

3. Satisfactory (D): fair but with significant shortcomings; it is issued on the basis of the arithmetic average of the three components of educational results (K, S, SC)

4. Good (C): generally sound work with a number of notable errors; it is issued on the basis of the arithmetic average of the three components of educational results (K, S, SC)

5. Very good (B): above the average standard but with some errors; it is issued on the basis of the arithmetic average of the three components of educational results (K, S, SC)

6. Excellent (A): outstanding performance with only minor errors; it is issued on the basis of the arithmetic average of the three components of educational results (K, S, SC)
